Objective: To establish the HR-MS-database of macrocyclic dilactone skeleton alkaloids from Tripterygium, and apply it in the fast identification and analysis of the alkaloids in Tripterygium form three Tripterygium species. Methods: Ultra-high performance liquid chromatography-quadrupole-time-of-flight mass spectrometry(UPLC/Q-TOF-MS)and LibraryView of SCIEX were used to construct the HR-MS-database of macrocyclic dilactone skeleton alkaloids in Tripterygium. The further research was performed for differences of the alkaloids in samples from three species of Tripterygium genus. Results: The HR-MS-database has been constructed, including 65 kinds of alkaloids in Tripterygium. The determination results of multiple samples from different habitats showed that the chemical composition of alkaloids in Tripterygium were significantly different, which contained 35 kinds of ingredients. And Tripterygium regelii Sprague et Takeda varied significantly from Tripterygium hypoglaucum (Levl.) Hutch and Tripterygium wilfordii Hook. f. Conclusion: The study provides an effective method for fast identification of alkaloids in Tripterygium with the macrocyclic dilactone skeleton. It is indicated that there are significant difference in alkaloids between different Tripterygium species. And it also provides reference for investigation on quality evaluation and substance basis of Tripterygium.
