Objective: To investigate the bioavailability of cadmium(Cd) in Oldenlandia diffusa Willd. in order to scientifically evaluate the health risk of Cd and provide basis for the establishment of limit standard. Methods: Inductively coupled plasma mass spectrometry (ICP-MS) was applied to determine the contents of Cd in Oldenlandia diffusa. The in vitro physiologically based extraction test(PBET) digestion/Caco-2 cell model was established to investigate the bioavailability of Cd in Oldenlandia diffusa. Furthermore, the hazard index method(HI method) was used to assess the risks of total contents and the bioavailable contents of Cd in Oldenlandia diffusa. Results: The total contents of Cd in three batches of Oldenlandia diffusa ranged from10.970 to 81.492 mg·kg-1. After taken up by Caco-2 cells, the bioavailability of Cd in Oldenlandia diffusa was 3.06%, 1.60%, and 6.62%, respectively. The results of risk assessment revealed that before taken up by Caco-2 cells, HI values of Cd for all batches of Oldenlandia diffusa were greater than 1, which revealed that the risks of Cd was unacceptable. After taken up by Caco-2 cells, the HI values of Cd in three batches of Oldenlandia diffusa were≤1, which suggested that the risks were at the safe level. Conclusion: Cd in Oldenlandia diffusa may not be totally absorbed by human body. This study provided the vital technical support for a more scientific and objective assessment of the health risks of heavy metals in traditional Chinese medicines, and for a more scientific and reasonable limit standard of heavy metals.
