Fingerprint analysis of Sophorae Fructus Carbonisata by linear calibration using two reference substances*

Abstract

Objective: To establish the HPLC fingerprints of Sophorae Fructus Carbonisata with analysis using the linear calibration using two reference substances method. Methods: The fingerprints of Sophorae Fructus Carbonisata were established by HPLC, and similarity analysis was used for quality evaluation. Linear calibration using two reference substances was used to identification of chromatographic peaks. Results: Ten chromatographic peaks were selected as common peaks in the fingerprint of 18 batches of Sophorae Fructus Carbonisata, and a control characteristic chromatogram with rutin, genistin, kaempferol-3-O-rutinoside, sophorabioside, sophoricoside and genistein as characteristic peaks was established. Linear calibration using two reference substances was more accurate and suitable for more chromatographic columns than relative retention time method. Conclusion: Linear calibration using two reference substances can be used to assist the chromatographic peak identification, and is feasible, reliable and economical.
