Study on HPLC fingerprint chromatogram and quantitation method of seven coumarins in Cnidii Fructus decoction pieces*

Abstract

Objective: To establish an integrated analysis method of HPLC fingerprint, multi-component quantification and chemometrics, and to evaluate the quality attributes and differences of Cnidii Fructusdecoction pieces from different sources. Methods: Analysis was performed on a Thermo C18 column (250 mm×4.6 mm,5 μm) column with acetonitrile-water for gradient elution. The flow rate was 1.0 mL·min-1, the column temperature was 30 ℃, the injection volume was 10 μL, and the detection wavelength was 220 nm. Established HPLC fingerprints of Cnidii Fructus decoction pieces from different sources, and evaluated the quality of 25 batches of Cnidii Fructus decoction pieces combined with cluster analysis (CA), partial least squares-discriminant analysis (PLS-DA) and principal component analysis (PCA), the contents of seven coumarins components: xanthotol, bregaptol, xanthotoxin, bergaptent, imperatorin, osthole and isoimperatorin were determined simultaneously. Results: The established fingerprint method met the method requirements. The similarity of 25 batches of Cnidii Fructus decoction pieces was more than 0.90. A total of 19 peaks were calibrated and 5 of them were quantitative analyzed. The samples were divided into several categories according to different cultivation places through cluster analysis. The results of principal component analysis and cluster analysis were consistent. The quality of samples S16, S17 and S18 from Shandong province was better. The 7 coumarins of quantitative analysis, the linear relationship, precision, stability, repeatability, and sample recovery test results were all good, the content ranges of the above seven coumarins were 0-1.221 mg·mL-1, 0-0.724 mg·mL-1, 7.925-28.784 mg·mL-1, 15.330-39.118 mg·mL-1, 59.173-156.662 mg·mL-1, 74.108-189.682 mg·mL-1, 0-4.141 mg·mL-1 respectively, of which xanthotoxin, citronol and isoimperatorin were not tested in some batches. Conclusion: There are certain quality differences in the Cnidii Fructusdecoction pieces from different origins. The fingerprint qualitative and quantitative analysis of coumarins based on uniform chromatographic analysis conditions is convenient, accurate and reliable. It can provide reference for the quality standards and evaluation of Cnidii Fructus decoction pieces.
