Objective: To develop an analytical method with rhapontin as the marker for fake Rhei Radix et Rhizoma and its extract added in health food by UPLC-QTRAP-MS/MS. Methods: The sample was extracted by menthol with ultrasound extraction and then separated on Agilent Infinity Lab Poroshell 120 SB-C18 (100 mm×2.1 mm,2.7 μm) column with gradient elution. The mobile phase consisted of 5 mmol·L-1 HCOONH4 in water and acetonitrile. Negative MRM-IDA-EPI was used to quantify and identify the target. Results: The linear relationship was good in the range of 5-100 ng·mL-1, and the correlation coefficient (r2) was 0.998 7. The average recovery at 1.25 mg·kg-1, 2.5 mg·kg-1 and 12.5 mg·kg-1 levels were 86.3%-118.7% in five base materials including tea, capsule, tablet, granule and liquid. One of the 18 batches of the samples collected was detected fake Rhei Radix et Rhizoma and its extract. Conclusion: The method is accurate, sensitive, specific with low detection limit. It can be used for the qualitative and quantitative determination in slimming health foods.
