Determination of 44 kinds of dyestuffs in oxidative hair dye products by HPLC

Abstract

Objective: To establish a method for determination of 44 kinds of dyestuffs in oxidative hair dye products by HPLC. Methods: Samples were ultrasonically extracted by anhydrous ethanol with 2 g·L-1 sodium bisulfite solution (1∶1). Separation was performed on a Waters XBridge Shield RP18(250 mm×4.6 mm, 5 μm) column with the mobile phase of methanol and 0.005 mol·L-1 ammonium acetate solution (pH 7.7), and an Agilent ZORBAX SB Aq(250 mm×4.6 mm,5 μm)column with the mobile phase of methanol and 0.005 mol·L-1 potassium dihydrogen phosphate solution (pH 7.0) in gradient elution respectively. The flow rate was 1.0 mL·min-1, the column temperature was 30 ℃. The detection wavelengths were set at 230, 240, 250 nm. Results: The linear ranges of 44 kinds of dyestuffs showed good linearity (r>0.999 5),the limits detection (LOD) were in the range of 2-58 μg·g-1. The average recoveries of three different concentration levels were 80.5%-113.6% with the RSD of 0.10%-5.6%. The method was used to determine 20 batches of oxidized hair dye products. The results showed that no banned dyestuffs were detected, and 20 kinds of permissible dyestuffs were detected. The contents of these dyestuffs ranged from 0.002% to 1.53%, all of which did not exceed the maximum allowable concentration specified of Safety and Technical Standards for Cosmetics(2015 edition). Conclusion: The method has the characteristic of good operability,reproducibility and high sensitivity. It can be used to determine dyestuffs in oxidative hair dye products.
