Objective: To establish the quantitative analysis of multi-components by single marker (QAMS) method for the content determination of five flavonoids in Sphenomeris chinensis, and verify its accuracy and feasibility. Methods: The relative correction factors of orientin, glucosyl-vitexin, vitexin-2-o-rhamnoside and galuteolin were calculated by HPLC with vitexinas internal reference, and the contents of each component were calculated with the corresponding correction factors, so as to realize quantitative analysis of multi-components by single marker. At the same time, the contents of five flavonoids in sphenomeris chinensis weredetermined by external standard method, and the accuracy and feasibility of QAMSmethod were verified by comparing the results. Results: Within a certain linear range, the relative correction factors between vitexin and orientin, glucosyl-vitexin, vitexin-2-o-rhamnoside as well as galuteolin were 0.772, 1.368, 1.380 and 0.731, respectively. There was no significant difference betweenthe contents of 4 flavonoids in 27 batches of samples determined by two methods. Conclusion: The quantitative analysis of multi-components by single marker method for the contentdetermination of orientin, glucosyl-vitexin, vitexin-2-o-rhamnoside, vitexin and galuteolin is feasible, accurate and reliable, which can be used for quantitative analysis and quality control of sphenomeris chinensis and its decoction pieces.
