Objective: To establish a method for simultaneous determination of hydroxy safflower yellow A, gallic acid, ellagic acid, dehydrocostus lactone and costunolide in Mongolian medicine preparation Huricha-6 pills by quantitative analysis of multi-components by single marker (QAMS). Methods: The analysis was performed on a Shim-pack VP-ODS C18 column (250 mm×4.6 mm, 5 μm), with the mobile phase comprising of methanol (A)-acetonitrile (B)-0.5% phosphoric acid (C) solution flowing at 1.0 mL·min-1 by gradient elution. The column temperature was set at 25 ℃, and the detection wavelengths were set at 403 nm (hydroxy safflower yellow A), 271 nm (gallic acid), 254 nm (ellagic acid) and 225 nm (dehydrocostus lactone and costunolide). With hydroxy safflower yellow A as the internal reference substance, the relative correction factors(RCFs) of gallic acid, ellagic acid, dehydrocostus lactone and costunolide to hydroxy safflower yellow A were calculated and established. The RCFs were used for the contents analysis of Huricha-6 pills. Results: The standard curves of hydroxy safflower yellow A, gallic acid, ellagic acid, dehydrocostus lactone and costunolide showed good linearity in 24.57-196.58, 78.45-627.60, 101.03-808.2, 11.98-95.84 and 5.36-42.88 μg·mL-1, respectively. The average recoveries and the corresponding RSD were 98.6%(1.1%),99.7%(0.75%), 97.8%(1.4%),96.6%(1.1%) and 102.3%(1.2%), respectively. There were no significant differences between the results obtained by QAMS and those obtained by external standard method. Conclusion: The QAMS method is feasible and credible, and can be used for the multi-index quality evaluation of Mongolian medicine preparation Huricha-6 pills.
