Determination of four flavonoids in the roots of Millettia speciosa by ultra high performance liquid chromatography with solid phase extraction*

Abstract

Objective: To establish an UPLC method for the determination of liquiritigenin, isoliquiritigenin, formononetin and maackiain in the roots of Millettia speciosa. Methods: The samples were extracted by ultrasonic with 50% methanol aqueous and purified by polyamide solid phase extraction (SPE) column. The target compounds were separated on a BEH C18 chromatographic column(100 mm×2.1 mm, 1.7 μm) with a gradient elution of acetonitrile-water containing 0.1% acetic acid at the flow rate of 0.3 mL·min-1, the column temperature was 50 ℃, the injection volume was 3 μL, and the detection wavelength was set at 275 nm. Results: The linear ranges of liquiritigenin, isoliquiritigenin, formononetin and maackiain were 0.50-40.0 μg·mL-1, 1.75-140.0 μg·mL-1, 0.45-225.0 μg·mL-1, 1.42-570.0 μg·mL-1, respectively, and the average recoveries were 89.5%, 80.9%, 83.8% and 103.1%, respectively, with the RSD of 1.7%, 4.1%, 1.2% and 0.79%. The method was applied to the determination of the 4 flavonoids in 25 batches of the roots of Millettia speciosa from different areas. The contents of liquiritigenin, isoliquiritigenin, formononetin and maackiain were in the ranges of 0.56-14.26 μg·g-1, 1.25-43.27 μg·g-1, 0.33-59.56 μg·g-1, 4.97-294.94 μg·g-1, respectively. Conclusion: The established UPLC method is rapid, sensitive, accurate and can be used for the determination of flavonoids in the roots of Millettia speciosa, which can provide a reference for the quality control of this herbs.
