Simultaneous determination of seven components in Qiguiling mixture by LC-MS/MS*

Abstract

Objective: To establish an ultra performance liquid chromatography mass spectrometry (UPLC-MS/MS) method for the rapid determination of astragaloside Ⅳ, prim-O-glucosylcimifugin, 5-O-methylvisammioside, liquiritin, glycyrrhizic acid, hesperidin and saikosaponin a in Qiguiling mixture. Methods: The analysis was perfoemed on Kinetex C18 100A column (50 mm×2.1 mm, 2.6 μm), and the mobile phase was 0.1% formic acid aqueous solution (A)-5 mmol·L-1 ammonium formate methanol (B) in a gradient elution manner, the flow rate was 0.3 mL·min-1. Results: The 7 compents of astragaloside Ⅳ, prim-O-glucosylcimifugin, 5-O-methylvisammioside, liquiritin, glycyrrhizic acid, hesperidin and saikosaponin a had good linear relationships in the range of 0.097 98-4.899, 0.031 64-1.582, 0.032 12-1.606, 0.052-2.6, 0.060 75-3.037 5, 0.306 54~15.327, 0.024~1.2 mg·L-1, and the average recoveries were between 95.4% and 105.0%. The contents of 7 components in 4 batches of Qiguiling mixture were 1.074 4-1.104, 0.279 9-0.288 7, 0.519 5-0.545 5, 0.437 8-0.478 6, 0.570 3-0.654 3, 1.123 4-1.363 6, 0.567 3-0.597 4 g·L-1, respectively. Conclusion: The method is rapid, sensitive and accurate, which can provide a reference for the quality evaluation of Qiguiling mixture.
