Identification study on Morindae Officinalis Radix decoction pieces and the fakes

Abstract

Objective: To distinguish Morindae Officinalis Radix decoction pieces and the fakes based on DNA barcoding technology, traditional identification and physicochemical identification technology. Methods: Internal transcribed spacer 2(ITS2) sequences of Morindae Officinalis Radix decoction pieces and the fakes were amplified with polymerase chain reaction(PCR).Through the method of BLAST comparison with GenBank database and establishment of NJ Tree, the origin of Morindae Officinalis Radix decoction pieces and the fakes was identified.The intra-species and inter-species differences among Morindae Officinalis Radix decoction pieces and the fakes were compared. And the differences among Morindae Officinalis Radix decoction pieces and the fakes in the properties of pharmacognosy characteristics and TLC behavior were compared. Results: The results of BLAST comparison with GenBank database and establishment of NJ Tree showed that Morindae Officinalis Radix decoction pieces and the fakes had obvious branches, and the 11 batches of fakes were identified as 7 species, including Morinda umbellata, Mitchella undulata, Ardisia crenada, Akebia quinata, Kadsura coccinea, Schisandra propinqua and Kadsura longipedunculata. Conclusion: Morindae Officinalis Radix decoction pieces and the fakescan be distinguished effectively based on the method of DNA barcoding technology. It provides a basis for the identification and standard improvement of Morindae Officinalis decoction pieces.
