Objective: To establish an HPLC-MS/MS method using acid assisted microwave hydrolysis for the quantitation of total L-carnitine in slimming health foods. Methods: The total L-carnitine in sample was extracted by acid assisted microwave hydrolysis after the sample was dissolved in water. After diluted,the solution was injected and separation was performed on Agilent Zorbax 300-SCX(3.0 mm×50 mm,1.8 μm) column. Water- acetonitrile(1∶1) containing 0.2% fomic acid with ammonium formate were used as mobile phase by gradient elution. The analyte was detected by positive ion ESI in Multi-reaction detection( MRM),with m/z 162.1→ 103.0 as the quantitative ion pair and m/z 162.1 → 60.2 as the qualitative ion pair. Results: The linearity of L-carnitine ranged from 10 ng·mL-1 to 5 000 ng·mL-1 with the correlation coefficient of 0.999 9. The limits of quantitation were 0.005 mg·g-1 in solid preparations( tablets,capsules) and 0.000 5 mg·g-1 in liquid preparations( oral liquid),respectively. Average recoveries under the 5 added concentrations were 91.6%-103.3%,and RSDs( n=6) were 0.83%-3.8%. The contents of L-carnitine in L-carnitine lotus leaf Alisma effervescent tablets,oyster soybean peptide carnitine oral liquid and L-carnitine green tea capsules were 0.174 g·g-1,0.833 mg·mL-1 and 18.78 mg·g-1,respectively. Conclusion: This method is suitable for the determination of L-carnitine in slimming health foods.
