Simultaneous determination of the six main components in Congrong Shujing granules by HPLC with multi-wavelength switching*

Abstract

Objective: To establish an HPLC multi-wavelength switching method for the simultaneous determination of echinacoside,paeonia lactiflora,verbascoside,salvianolic acid B,paeonol,and tanshinone ⅡA in Congrong Shujing granules. Methods: An Agilent Zorbax Eclipse XDB-C18column(250 mm×4.6 mm,5 μm) was used, with acetonitrile-0.1% phosphoric acid solution as the mobile phase in gradient elution. The flow rate was 1.0 mL·min-1,the column temperature was 25 ℃,the injection volume was 20 μL,and the detection wavelengths were set at 330 nm(echinacoside and verbascoside),230 nm(paeoniflorin),286 nm(salvianolic acid B and paeonol) and 270 nm(tanshinone ⅡA). Results: Echinacoside,paeoniflorin,verbascoside,salvianolic acid B,paeonol and tanshinone ⅡA showed good linearity in the ranges of 18.18-181.8 μg·mL-1r=0.999 8),28.86-288.6 μg·mL-(1 r=0.999 8),0.576 0-5.760 μg·mL-(1 r=0.999 5),32.96-329.6 μg·mL-(1 r=0.999 7),31.97-319.7 μg·mL-(1 r=0.999 0),1.120-11.20 μg·mL-(1 r=0.999 8),respectively . The average recovery(n=6) were 98.7%, 99.4%,98.6%,99.0%,98.8%,99.1% with RSDs of 1.0%,0.92%,1.6%,0.85%,1.0%,and 1.7%,respectively. The content ranges of the above-mentioned components in 3 samples were 3.571-3.702 mg·g-1,6.692-6.911 mg·g-1,0.071-0.089 mg·g-1,7.343-7.947 mg·g-1,2.201-2.259 mg·g-1 and 0.041-0.055 mg·g-1,respectively. Conclusion: This method can be used as the quality standard of Congrong Shujing granules to simultaneously determine the contents of six main components.
