HPLC fingerprint and chemical pattern recognition of Xingpi Yang’er granules

Abstract

Objective: To establish an HPLC chromatographic fingerprint of Xingpi Yang’er granules. Methods: The separation was performed on an Agilent TC- C18 column(250 mm×4.6 mm,5 μm)with the mobile phase consisting of acetonitrile and 0.05% phosphoric acid(gradient elution)at a flow rate of 1.0 mL·min-1. The detection wavelength were 210,282 and 350 nm,the column temperature was 30 ℃ and the injection volume was 10 μL. Similarty evaluation software combined with principal component analysis(PCA)and discriminant analysis of partial least squares(PLS-DA)were used to evaluate the HPLC fingerprints of 14 batches of Xingpi Yang’er granules. Results: There were 19 common peaks in the HPLC fingerprints of 14 batches of Xingpi Yang’er granules,and 7 common components were identified. The similarities of 14 batches of samples were 0.983- 0.997. Totally 14 batches of samples were classified into three groups and a brief list of four different markers was presented by PCA and PLS-DA. Conclusion: This method is simple,stable and repeatable,providing experimental basis for qualitative and quantitative analysis of Xingpi Yang’er granules.
