目的:建立肉苁蓉、管花肉苁蓉及沙苁蓉中松果菊苷、管花苷 A、毛蕊花糖苷、异毛蕊花糖苷、金石 蚕苷和 2′-乙酰金石蚕苷的 UPLC 含量测定方法,并比较其含量的异同。方法:采用 Waters HSS T3(100 mm×2.1 mm,1.8 μm)色谱柱,流动相为甲醇 -0.1% 甲酸水,梯度洗脱,流速 0.3 mL·min-1,检测波长 330 nm,柱温 30 ℃,进样量 1 μL。获得数据进行化学计量学评价,并对 6 个成分的含量测定进行方法学考 察。结果:在上述条件下,6 个成分均获得良好的分离,其线性范围分别为 30.80~1 232、0. 798 0~31.91、 3.310~132.4、0.933~37.32、59.02~885.3、35.73~536.0 μg·mL-1,线性关系良好(r ≥ 0.999 9);平均加样回收率(n=6)分别为 98.5%、97.3%、98.7%、99.3%、99.7% 和 98.4%;精密度、重复性和稳定性的 RSD 均 <4.0%。金石蚕苷、2′-乙酰金石蚕苷为沙苁蓉特有成分,毛蕊花糖苷为肉苁蓉、管花肉苁蓉及沙苁蓉共有成分但种间差异大,沙苁蓉所含苯乙醇苷的种类和含量与肉苁蓉及管花肉苁蓉均有较大的差异;主成分分析可将正品肉苁蓉与伪品沙苁蓉明显区分开。结论:该方法简便,准确,重复性好,可为肉苁蓉、管花肉苁蓉及沙苁蓉的质量评价及种间鉴别提供参考依据。
Objective:To establish a UPLC method for the determination of echinacoside,cistanoside A, acteoside,isoacteoside,poliumoside and 2′-acetylpoliumoside in Cistanche deserticola,Cistanche tubulosa and Cistanche sinensis,and compare their contents. Methods:UPLC analysis was performed on Waters HSS T3(2.1 mm×100 mm,1.8 μm)with a mobile phase consisting of methanol -0.1% formic acid aqueous with gradient eluent at a flow rate of 0.3 mL·min-1 and the injection volume was 1μL. The detection wavelength was 330 nm and the column temperature was 30 ℃ . The data were evaluated by chemometrics. The six components were determined. Results:Under the above conditions,the linear relationship for six components were separated well and in the ranges of 30.80-1 232 μg·mL -1,0. 798 0-31.91 μg·mL -1,3.310-132.4 μg·mL -1,0.933-37.32 μg·mL-1,59.02-885.3 μg·mL -1 and 35.73-536.0 μg·mL -1,respectively,with good linear relationship r ≥ 0.999 9. The average recoveries(n=6)were 98.5%,97.3%,98.7%,99.3%,99.7% and 98.4%,respectively. The RSDs of precision,repeatability and stability tests were < 4.0%. Poliumoside and 2’-acetylpoliumoside were the specific components of Cistanche sinensis,while acteoside was the common component of Cistanche deserticola,Cistanche tubulosa and Cistanche sinensis,but the species and content of phenylethanolic glycosides in Cistanche sinensis were different from those of Cistanche deserticola and Cistanche tubulosa. The principal component analysis could distinguish the genuine Cistanche deserticola from the fake Cistanche sinensis. Conclusion:The method is simple, accurate and reproducible,which can provide reference for quality evaluation and interspecific identification of Cistanche deserticola,Cistanche tubulosa and Cistanche sinensis.
