Comparative studies of four Cistanche speices based on HPLC characteristic chromatogram

摘要

目的: 采用高效液相色谱法建立荒漠肉苁蓉、沙苁蓉药材的特征图谱, 对不同基原肉苁蓉进行鉴别研究, 为同属药材的鉴定及质量评价提供参考。方法: 采用Agilent zobarx C18 色谱柱(250 mm×4. 6 mm, 5 μm), 乙腈(A)-0. 1% 甲酸水溶液为流动相, 梯度洗脱, 流速1. 0 mL·min-1, 检测波长237 nm, 柱温35 ℃, 采用中药色谱指纹图谱相似度评价软件(2012 年版)对4 种肉苁蓉进行相似度分析。结果: 沙苁蓉与荒漠肉苁蓉、管花肉苁蓉和盐生肉苁蓉所含化学成分存在明显不同。其中标定了荒漠肉苁蓉、管花肉苁蓉和盐生肉苁蓉10 个共有峰, 鉴定其中7 个共有峰;标定沙苁蓉10 个共有峰, 鉴定其中5 个共有峰。通过比较不同基原肉苁蓉样品的相似度, 可以准确区分荒漠肉苁蓉和沙苁蓉。进一步计算肉苁蓉苷A 与松果菊苷、2'-乙酰毛蕊花糖苷与毛蕊花糖苷的峰面积比值, 比值在荒漠肉苁蓉和管花肉苁蓉中差异显著, 可用于药材品种的鉴定。结论: 本研究所建立的荒漠肉苁蓉、沙苁蓉特征图谱专属性强, 可以准确区分不同基原肉苁蓉, 为规范苁蓉类药材的使用提供参考, 同时为全面控制和评价肉苁蓉属药材质量提供科学依据。

Abstract

Objective: To establish HPLC characteristic chromatograms of Cistanche deserticola and Cistanche sinensis and to identify different Cistanche speices, so as to provide references for the identification and quality evaluation of the herb. Methods: HPLC analysis was performed on an Agilent Zobarx C18 column(250 mm×4. 6 mm, 5 μm). The mobile phase consisted of acetonitrile(A)and 0. 1% formic acid aqueous solution(B)with gradient elution. The flow rate was 1 mL·min-1, the detection wavelength was 237 nm and the column temperature was 35 ℃. TCM chromatographic fingerprint similarity evaluation software(2012 version)was used for similarity analysis of four cistanche speices. Results: The chemical components of sinensis were different from those of C. deserticola, C. tubulosa and C. salsa. Among them, 10 common peaks of C. deserticola, C. tubulosa and C. salsa were marked, and 7 of them were identified. Ten common peaks of C. sinensis were marked and 5 of them were identified. By similarity evaluation software, the similarities of four Cistanche speices were compared. There were obvious differences between the characteristic chromatograms of C. deserticola and C. sinensis. There was no significant difference among the similarities of C. deserticola, C. tubulosa and C. salsa. On this basis, the peak area ratio of cistanoside A and echinacoside, and the peak area ratio of 2'-acetylacteoside and acteoside can be used as reference indexes to distinguish C. deserticola and C. tubulosa. Conclusion: The new method is suitable for the identification of C. deserticola, C. tubulosa and C. sinensis with high efficiency specificity. It provides a scientific basis for the identification and quality evaluation of them.
