摘要

目的: 蛤蚧为我国的名贵中药材,建立PCR-核酸试纸条检测方法对蛤蚧进行定性鉴别。方法: 通过碱裂解法提取蛤蚧及其伪品的基因组DNA,以蛤蚧细胞色素C氧化酶亚基Ⅰ(CoⅠ)为靶基因,设计特异性引物,确定合适的PCR-核酸试纸条反应体系以及反应条件。结果: 通过PCR-核酸试纸条对蛤蚧进行定性分析,正品蛤蚧在PCR-核酸试纸条上均出现2条带,伪品以及阴性对照均出现1条带。结论: PCR-核酸试纸条检测方法可实现短时间内蛤蚧可视化检测,操作方便,结果准确,可以为蛤蚧的DNA分子鉴别提供技术保障。

Abstract

Objective: To establish a PCR-nucleic acid test strip method to identify the authenticity of Gekko gecko, which is a precious Chinese medicinal herb in China,. Methods: The genomic DNA of Gekko gecko and its adulterants was extracted by alkaline denaturation method. With cytochrome C oxidase subunit I (CoI) as the target gene, specific primers were designed to determine the appropriate PCR-nucleic acid test strip reaction system and reaction conditions. Results: In this experiment, the qualitative analysis of Gekko gecko was carried out by PCR-nucleic acid test strip. The authentic Gekko gecko showed two bands on the PCR-nucleic acid test strip, while the counterfeit Gekko gecko and negative control both showed one band. Conclusion: The PCR-nucleic acid test strip detection method can achieve the visual detection of Gekko gecko in a short time, with wide application range, simple operation and accurate results, which can provide technical support for the identification of Gekko gecko DNA molecules.
