摘要

目的: 建立龙骨安神酒HPLC特征图谱并测定斯皮诺素、金丝桃苷、朝藿定A、朝藿定B、朝藿定C、淫羊藿苷、补骨脂素、异补骨脂素、宝藿苷Ⅰ、五味子醇甲的含量,为龙骨安神酒的质量控制提供参考。方法: 采用Agilent Zorbax SB-C18色谱柱(4.6 mm×250 mm,5 μm),以乙腈-水溶液为流动相进行梯度洗脱,体积流量为1.0 mL·min-1,柱温为30 ℃,检测波长为270 nm;通过相似度评价,结合聚类分析(CA)、主成分分析(PCA)及偏最小二乘法-判别分析(PLS-DA)对12批龙骨安神酒的指纹图谱进行分析评价,并对指认的10个指标成分进行定量测定研究。结果: 建立了龙骨安神酒特征指纹图谱,共标定了15个共有峰,各批次样品的相似度均大于0.9;12批龙骨安神酒样品质量存在一定的差异,筛选出对质量影响较大的5个关键成分,分别为斯皮诺素、金丝桃苷、朝藿定C、淫羊藿苷、补骨脂素; 10种成分的线性关系良好(r≥0.999 6),含量范围分别为10.59~18.59、48.51~61.11、13.74~20.78、13.05~20.05、231.82~271.29、113.77~148.98、60.89~80.58、58.90~71.68、17.91~21.17、64.09~76.34 μg·mL-1结论: 建立的龙骨安神酒特征图谱及10个成分含量测定的方法,简便、准确可靠,可用于龙骨安神酒的质量控制。

Abstract

Objective: To establish a method for charateristic chromatogram of Longgu Anshen liquor and to determine the contents of spinosin, hypericin, epimedin A, epimedin B, epimedin C, icariin, psoralen, isopsoralen, baohuosideⅠand schisandra A, and provide reference its quality control. Methods: Agilent Technologies Zorbax SB-C18 column (4.6 mm×250 mm, 5 μm) was used, with the mobile phase comprising of acetonitrile-water flowing at 1.0 mL·min-1 in a gradient elution manner,the column temperature was 30 ℃,and the detection wavelength was set at 270 nm. Based on similarity evaluation, cluster analysis (CA), principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA), the fingerprints of 12 batches of Longgu Anshen liquor were analyzed and evaluated, and the 10 index components identified were quantitatively determined. Results: The HPLC fingerprint of Longgu Anshen liquor was constructed with 15 common chromatographic peaks, The similarities of each batches of samples were all above 0.9 comparing with the reference. There were some differences in the quality of 12 batches of Longgu Anshen liquor samples, and five key components were screened out, which were spinolin, hyperin, epimedin C, icariin and psoralen.The 10 constituents showed good linear relationship(r≥0.999 6), whose content ranges were 10.59-18.59 μg·mL-1,48.51-61.11 μg·mL-1,13.74-20.78 μg·mL-1,13.05-20.05μg·mL-1,231.82-271.29 μg·mL-1,113.77-148.98 μg·mL-1,60.89-80.58 μg·mL-1,58.90-71.68 μg·mL-1,17.91-21.17 μg·mL-1,64.09-76.34 μg·mL-1. Conclusion: The established charateristic chromatograms of Longgu Anshen liquor and the content determination method, simple accurate and reliable,and can be used for its quality control.
