摘要

目的: 应用简单重复序列(SSR)标记对人参属植物人参、西洋参和三七进行鉴别。方法: 利用EGassembler对NCBI的EST数据库进行预处理,SSRIT网站寻找SSR位点后用Primer3设计特异性引物。采用非变性聚丙烯酰胺凝胶电泳和毛细管电泳2种方法筛选并验证EST-SSR引物的通用性。应用筛选的SSR引物构建标准指纹图谱并对市售商业参类产品的原材料进行鉴定。结果: 应用生物信息学的方法共筛选出7对EST-SSR标记可以用于人参、西洋参和三七的鉴定。在市售商业产品的检测中,以引物P2和Q11的联合检测阳性率最高。结论: SSR标记具有较高的重复性和准确性,可以作为人参属植物可靠的分子标记。

Abstract

Objective: Using the simple sequence repeat (SSR) markers to identify Panax ginseng, Panax quinquefolium and Panax notoginseng. Methods: The EST database of NCBI was pre-processed by using EGassembler. SSRIT website was used to identify SSR loci and Primer3 was used to design specific primers. The universality and polymorphism of EST-SSR primers were screened and verified by non-denatured polyacrylamide gel electrophoresis and capillary electrophoresis. These SSR primers were used to construct standard fingerprints and identify the raw materials of ginseng products from the market. Results: Through bioinformatics methods, seven pairs of EST-SSR markers were screened for identification of Panax ginseng, Panax quinquefolium and Panax notoginseng. The combination of primers P2 and Q11 showed the highest positive rate in the detection of commercial products. Conclusion: SSR molecular markers are repeatable and accurate, which can be used as reliable molecular markers for identification of Panax species.
