目的:明确驱虫斑鸠菊药材的基原、鉴别要点以及DNA条形码序列信息。方法:应用体式荧光显微镜、光学显微镜和扫描电镜及其数字化成像技术, 对驱虫斑鸠菊进行性状和显微鉴定研究, 并应用DNA 条形码技术进行分子生物学鉴定研究。结果:驱虫斑鸠菊果实在外形、棱肋、肋间、被毛等性状鉴定特征, 以及石细胞群和纤维群、棱脊处维管束、中果皮内侧“V”字形石细胞带、草酸钙柱晶和方晶等显微鉴定特征上具有明显的鉴别意义。16批驱虫斑鸠菊药材的DNA 条形码鉴定结果一致, 其ITS和ITS2序列在GenBank数据库中均可比对到驱虫斑鸠菊Vernonia anthelmintica, 且一致性为100%。结论:明确了驱虫斑鸠菊药材性状鉴定、显微鉴定的要点, 以及ITS、ITS2序列的信息, 为驱虫斑鸠菊药材的性状鉴定、显微鉴定、DNA条形码鉴定提供依据。

Objective:To clarify the origin, identification points and DNA barcode sequence information of Fructus Vernoniae. Methods:The macroscopic and microscopic identification of Fructus Vernoniae was studied using stereo microscopy, optical microscopy and scanning electron microscopy combined with digital imaging technology. And the molecular identification of this drug was studied using DNA barcoding technology. Results:The characteristics in appearance, ribs, intercostals, coats, etc of Vernonia anthelmintica showed obvious distinguishing significance in stone cell clusters, fiber clusters, vascular bundles at the ridges, “V”-shaped stone cell belts on the inner side of the mesocarp, calcium oxalate columnar crystals and squares features such as crystal. The DNA barcode identification results of 16 batches of Fructus Vernoniae medicinal materials were consistent, its ITS and ITS2 sequences could be compared to Vernonia anthelmintica in the GenBank database, and the consistency was 100%. Conclusion:The main points of the macroscopic and microscopic identification of Fructus Vernoniae, and the information of ITS, ITS2 sequences, which providing basis for the character identification, microscopic identification, and DNA barcode identification of Fructus Vernoniae is clarified.
