目的:基于Taqman探针结合熔解曲线技术, 建立1种简单、高效的快速检测抗高血压药物相关基因多态位点的基因分型方法, 指导临床上抗高血压药物的个性化给药方式, 从而有效降低不良反应发生率。方法:随机采集200例原发性高血压患者的口腔粘膜脱落细胞, 通过提取口腔粘膜脱落细胞基因组DNA, 设计特异性的Taqman探针和不对称扩增引物, 在2个反应管中采用多色Taqman探针结合熔解曲线技术对提取的基因组DNA的5个抗高血压药物相关基因多态位点进行基因分型检测, 并随机抽取不同基因型的样本进行基因测序验证。结果:Taqman探针结合熔解曲线技术能够对200例抽提的基因组DNA的5个抗高血压药物相关基因多态位点进行准确的分型, 分型结果与基因测序结果也完全一致。结论:Taqman探针熔解曲线技术能够有效的对复杂样本类型的基因组DNA 进行基因分型, 是1种简单、快速、精确的检测方法, 可以适用于多种突变类型的检测。
