目的: 基于高分辨质谱多肽鉴定技术分析脾氨肽中多肽类物质,并采用生物信息技术预测多肽的生物活性。方法: 以Orbitrap Fusion Lumos三合一高分辨质谱系统对混合多肽进行鉴定。利用BIOPEP-UWM数据库工具预测所有肽段的潜在生物活性。结果: 脾氨肽中鉴定出1 461条肽段,分别来源于198种前体蛋白,主要有血红蛋白、肌动蛋白、组蛋白、胸腺素β4等。肽段片段被预测具有53种生物活性类型,涵盖免疫调节、免疫刺激及抗炎等功能。结论: 超高效液相-高分辨质谱联合技术及生物信息技术为多组分生化药分析鉴定及活性预测提供便捷手段。脾氨肽免疫调节作用的物质基础可能为免疫相关活性多肽。
Objective: To analyze the peptides in spleen aminopeptide using high-resolution mass spectrometry and to predict their bioactivity using bioinformatics technology. Methods: The mixed peptides were identified using the Orbitrap Fusion Lumos high-resolution mass spectrometry system. BIOPEP-UWM database tool was employed to predicted the potential biological activities of all peptides. Results: 1 461 peptides were identified from 198 precursor proteins, such as hemoglobin, actinin, histone, thymosin β4, etc. The peptide fragment is predicted to possess 53 types of biological activities, including immunomodulating, immunostimulating, and anti-inflammatory, etc. Conclusion: The combination of ultra-high-performance liquid chromatography and high-resolution mass spectrometry with bioinformatics provides a convenient approach for the analysis and prediction of the activities of multicomponent biochemical drugs. The immunomodulatory effects of spleen aminopeptides may be attributed to the direct immune activities of the peptides as well as the combined effects of various biological activities related to immunity.
