Study on extraction process and antifungal activity of ethanol extract from leaves of Cinnamomum camphora Chvar. Camphor*

Abstract

Objective: To explore the optimal extraction process of its alcohol extract, and the antibacterial activity and possible chemical components of petroleum ether, ethyl acetate and n-butanol extraction phases, while the leaves of Cinnamomum camphora Chvar. camphor were used as raw materials. Methods: The thermal reflux extraction process of the alcohol extracts from leaves of Cinnamomum camphora Chvar. camphor was optimized by using single-factor experiments combined with the response surface method. The antifungal activities of different extracts against F. oxysporum, C. gloeosporioides, T. cucumeris and F. solani were studied by using the method of toxic plate and mycelial growth rate; The chemical constituents of different extraction phases were detected and analyzed by LC-MS. Results: The optimum extraction conditions were as follows: 95% ethanol, solid-liquid ratio 1 ∶ 25 (g · mL-1), extraction time 3 h, and the extraction rate was 31.29%. Different extraction phases of the ethanol extract of leaves of Cinnamomum camphora Chvar. camphor had inhibitory effects on the tested strains. When the concentration of the extraction phase was 0.5%, the petroleum ether extraction phase had the strongest antibacterial ability against C.gloeosporioides, and the antifungal rate was (72.82±3.20)%. Ethyl acetate and n-butanol extraction had significant antibacterial effect on R. solani, and the antibacterial rate was (100.00±0.00)%, which could completely inhibit its growth. In addition, it was preliminarily speculated that the ethanol extract contained at least 17 compounds. Conclusion: The optimized extraction process is stable and feasible, the antibacterial effect of different extraction phases on the tested bacteria is clarified, and the compounds contained in the camphor-type alcohol extract are preliminarily speculated, This laid the foundation for the application of camphor trees in the field of natural antibacterial agents.
